I want to set up a LAN in my home between two computers and I have a Netgear WGR614 router, but I have a dial-up connection. Can I set up the LAN without an RJ-45 cable connection or is this absolutely necessary?

Yes, you need a UTP RJ-45 cable. Unless you have an older coax connection on the PCs and router  :D

Wireless is also an option, but you'd need a new router or access point (which, now that I'm thinking about it, will also need an RJ-45 cable).

Now, check out www.Easy-Tek.com (http://www.Easy-Tek.com) if have any other problems :D You can find me there as Greg H .

EDIT: ah just, you've got dial-up? Well then, as cybermonk says below, you don't even need the router. But you will still need a RJ-45 cable crossover cable. Or two wireless cards if you want to use that method...

If its only two computers you dont need a router, just throw an ethernet line between the two. Also, I dont think whether you have a dial up or cable or dsl line has any effect on the ability to set up a lan. Hope it helps.

Without the router, I couldn't get the ethernet connection to work...
The computers seemed connected, but they couldn't really communicate or anything. With the router installed, now only one computer can view the other's shared files, but that is about all I can get it to do.
